Background/Introduction: Human BD-2, also known as Beta-Defensin 2, is a 64 amino acid cytokine protein encoded by the DEFB2 gene. After signal sequence processing and maturation, the cytokine becomes 41 amino acid residues long (from 24 to 64). Defensins form a family of microbicidal and cytotoxic peptides made by neutrophils. Members of the defensin family are highly similar in protein sequence. Moreover, it has been recently determined that Human BD-2 itself is a cysteine-rich antimicrobial cytokine present on lesional skin. The cytokine is produced by a variety of epithelial cells and exhibits potent antimicrobial activity against Gram-negative but not Gram-positive bacteria, thereby minimizing the frequency of Gram-negative infections on skin and lung tissue. Human BD-2 is the first defensin implicated following stimulation of epithelial cells by contact with microorganisms such as Pseudomonas Aeruginosa or other cytokines such as TNF-alpha or IL-1beta. Typically expressed in keratinocytes associated with inflammatory skin lesions, hBD-2 is a dynamic component of epithelial defense systems that protect skin and respiratory tract surfaces from bacterial infection.
