MyBioSource MBS822016 image 2
Immunohistochemical analysis of Cytochrome P450 2J2 staining in human liver cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

Immunofluorescent analysis of Cytochrome P450 2J2 staining in A549 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

ncbi summary :
This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum and is thought to be the predominant enzyme responsible for epoxidation of endogenous arachidonic acid in cardiac tissue. [provided by RefSeq, Jul 2008]
uniprot summary :
CYP2J2: This enzyme metabolizes arachidonic acid predominantly via a NADPH-dependent olefin epoxidation to all four regioisomeric cis-epoxyeicosatrienoic acids. One of the predominant enzymes responsible for the epoxidation of endogenous cardiac arachidonic acid pools. Belongs to the cytochrome P450 family. Protein type: Oxidoreductase; Lipid Metabolism - arachidonic acid; Lipid Metabolism - linoleic acid; EC 1.14.14.1; Contractile. Chromosomal Location of Human Ortholog: 1p31.3-p31.2. Cellular Component: endoplasmic reticulum membrane; intracellular membrane-bound organelle; cytoplasm. Molecular Function: arachidonic acid 14,15-epoxygenase activity; iron ion binding; heme binding; arachidonic acid 11,12-epoxygenase activity; oxidoreductase activity, acting on paired donors, with incorporation or reduction of molecular oxygen, reduced flavin or flavoprotein as one donor, and incorporation of one atom of oxygen; arachidonic acid epoxygenase activity; steroid hydroxylase activity; oxygen binding. Biological Process: xenobiotic metabolic process; exogenous drug catabolic process; icosanoid metabolic process; arachidonic acid metabolic process; linoleic acid metabolic process; epoxygenase P450 pathway; regulation of heart contraction
