Western Blot (WB), Immunofluorescence (IF), Immunocytochemsitry (ICC)

WB (1/500 - 1/1000), IF/ICC (1/100 - 1/500). Western blot analysis of 14-3-3 epsilon expression in KNRK (A), SW480 (B), Caco2 (C), NIH3T3 (D), H9C2 (E) whole cell lysates. Immunofluorescent analysis of 14-3-3 epsilon staining in NIH3T3 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 degree C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

Immunogen: KLH-conjugated synthetic peptide encompassing a sequence within the C-term region of human 14-3-3 epsilon. The exact sequence is proprietary.

This gene product belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 100% identical to the mouse ortholog. It interacts with CDC25 phosphatases, RAF1 and IRS1 proteins, suggesting its role in diverse biochemical activities related to signal transduction, such as cell division and regulation of insulin sensitivity. It has also been implicated in the pathogenesis of small cell lung cancer. Two transcript variants, one protein-coding and the other non-protein-coding, have been found for this gene.
