WB (1/500 - 1/1000), IF/ICC (1/100 - 1/500). Western blot analysis of Histamine H2 Receptor expression in HEK293T (A), COLO205 (B), NIH3T3 (C), H9C2 (D) whole cell lysates. Immunofluorescent analysis of Histamine H2 Receptor staining in COLO205 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 degree C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

ncbi summary :
Histamine is a ubiquitous messenger molecule released from mast cells, enterochromaffin-like cells, and neurons. Its various actions are mediated by histamine receptors H1, H2, H3 and H4. Histamine receptor H2 belongs to the family 1 of G protein-coupled receptors. It is an integral membrane protein and stimulates gastric acid secretion. It also regulates gastrointestinal motility and intestinal secretion and is thought to be involved in regulating cell growth and differentiation. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Aug 2008]
uniprot summary :
H2R: The H2 subclass of histamine receptors mediates gastric acid secretion. Also appears to regulate gastrointestinal motility and intestinal secretion. Possible role in regulating cell growth and differentiation. The activity of this receptor is mediated by G proteins which activate adenylyl cyclase and, through a separate G protein-dependent mechanism, the phosphoinositide/protein kinase (PKC) signaling pathway. Belongs to the G-protein coupled receptor 1 family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Membrane protein, multi-pass; GPCR, family 1; Receptor, GPCR; Membrane protein, integral. Chromosomal Location of Human Ortholog: 5q35.2. Cellular Component: integral to plasma membrane; plasma membrane. Molecular Function: histamine receptor activity. Biological Process: gland development; G-protein signaling, coupled to cyclic nucleotide second messenger; gastrin-induced gastric acid secretion; gut development; histamine-induced gastric acid secretion; positive regulation of vasoconstriction; visual learning; regulation of synaptic plasticity; immune response; memory
