other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Ctss were tested 20 times on one plate, respectively. Intra-Assay: CV<8%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Ctss were tested on 3 different plates, 8 replicates in each plate. CV (%) = SD/meanX100. Inter-Assay: CV<10%
products description :
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- Ctss antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- Ctss antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Ctss amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Ctss can be calculated.

ncbi summary :
The protein encoded by this gene, a member of the peptidase C1 family, is a lysosomal cysteine proteinase that may participate in the degradation of antigenic proteins to peptides for presentation on MHC class II molecules. The encoded protein can function as an elastase over a broad pH range in alveolar macrophages. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. uniprot summary :
CTSS: Thiol protease. Key protease responsible for the removal of the invariant chain from MHC class II molecules. The bond- specificity of this proteinase is in part similar to the specificities of cathepsin L and cathepsin N. Belongs to the peptidase C1 family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: EC 3.4.22.27; Protease. Chromosomal Location of Human Ortholog: 1q21. Cellular Component: extracellular region; extracellular space; intracellular membrane-bound organelle; lysosomal lumen; lysosome; membrane. Molecular Function: collagen binding; cysteine-type endopeptidase activity; fibronectin binding; laminin binding; proteoglycan binding. Biological Process: adaptive immune response; antigen processing and presentation; antigen processing and presentation of exogenous peptide antigen via MHC class I; antigen processing and presentation of exogenous peptide antigen via MHC class I, TAP-independent; antigen processing and presentation of exogenous peptide antigen via MHC class II; antigen processing and presentation of peptide antigen; antigen processing and presentation of peptide antigen via MHC class I; collagen catabolic process; extracellular matrix disassembly; extracellular matrix organization and biogenesis; immune response; innate immune response; positive regulation of inflammatory response; protein processing; proteolysis; proteolysis involved in cellular protein catabolic process; response to acidity; toll-like receptor signaling pathway
