Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level LELP1 were tested 20 times on one plate, respectively. Intra-Assay: CV<8%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level LELP1 were tested on 3 different plates, 8 replicates in each plate. CV (%) = SD/meanX100. Inter-Assay: CV<10%
products description :
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-LELP1 antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-LELP1 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the LELP1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of LELP1 can be calculated.

ncbi summary :
This gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in this gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. This gene has also been linked to type 2 diabetes mellitus development. [provided by RefSeq, Jul 2008]
uniprot summary :
leptin: May function as part of a signaling pathway that acts to regulate the size of the body fat depot. An increase in the level of LEP may act directly or indirectly on the CNS to inhibit food intake and/or regulate energy expenditure as part of a homeostatic mechanism to maintain constancy of the adipose mass. Defects in LEP may be a cause of obesity (OBESITY). It is a condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat. Belongs to the leptin family. Protein type: Hormone; Secreted; Cell development/differentiation; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 7q31.3. Cellular Component: cytoplasm; extracellular region; extracellular space. Molecular Function: growth factor activity; hormone activity; peptide hormone receptor binding.
