This assay has high sensitivity and excellent specificity for detection of TBA . No significant cross-reactivity or interference between TBA and analogues is observed. Note: Limited by current skills and knowledge, it is impossible for us to complete the crossreactivity detection between TBA and all the analogues, therefore, cross reaction may still exist.

Store at 4 degree C if kit is to be used within 1 week. Stable for 6 months (if micro ELISA Plate, Lyophilized Standard and Concentrated Biotinylated Detection Protein stored at-20 degree C. Other components at 2-8 degree C). Stable for 12 months (if the entire kit is stored at-20 degree C).

Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level TBA were tested 20 times on one plate, respectively. Intra-Assay: CV<8%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level TBA were tested on 3 different plates, 8 replicates in each plate. Principle of the Assay: This kit was based on Competitive-ELISA detection method. The microtiter plate provided in this kit has been pre-coated with TBA. During the reaction, TBA in the sample or standard competes with a fixed amount of TBA on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to TBA. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of TBA in the samples is then determined by comparing the O D of the samples to the standard curve.
