specificity :
This assay has high sensitivity and excellent specificity for detection of BTA. No significant cross-reactivity or interference between BTA and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between BTA and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 1.0 ng/mL.
other info2 :
Intended Uses: This BTA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human BTA. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
Principle of the assay: BTA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BTA antibody and an BTA-HRP conjugate. The assay sample and buffer are incubated together with BTA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BTA concentration since BTA from samples and BTA-HRP conjugate compete for the anti-BTA antibody binding site. Since the number of sites is limited, as more sites are occupied by BTA from the sample, fewer sites are left to bind BTA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BTA concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a member of the heparan sulfate/heparin GlcNAc N-deacetylase/ N-sulfotransferase family. The encoded enzyme is a type II transmembrane protein that resides in the Golgi apparatus. The encoded protein catalyzes the transfer of sulfate from 3'-phosphoadenosine 5'-phosphosulfate to nitrogen of glucosamine in heparan sulfate. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2014]
uniprot summary :
NDST1: Essential bifunctional enzyme that catalyzes both the N- deacetylation and the N-sulfation of glucosamine (GlcNAc) of the glycosaminoglycan in heparan sulfate. Modifies the GlcNAc-GlcA disaccharide repeating sugar backbone to make N-sulfated heparosan, a prerequisite substrate for later modifications in heparin biosynthesis. Plays a role in determining the extent and pattern of sulfation of heparan sulfate. Compared to other NDST enzymes, its presence is absolutely required. Participates in biosynthesis of heparan sulfate that can ultimately serve as L- selectin ligands, thereby playing a role in inflammatory response. Belongs to the sulfotransferase 1 family. NDST subfamily. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Hydrolase; EC 2.8.2.8; Glycan Metabolism - heparan sulfate biosynthesis; Transferase; Membrane protein, integral. Chromosomal Location of Human Ortholog: 5q33.1. Cellular Component: Golgi membrane; integral to membrane. Molecular Function: deacetylase activity; [heparan sulfate]-glucosamine N-sulfotransferase activity.
