. Principle of the assay: BMPR1B ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BMPR1B antibody and an BMPR1B-HRP conjugate. The assay sample and buffer are incubated together with BMPR1B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BMPR1B concentration since BMPR1B from samples and BMPR1B-HRP conjugate compete for the anti-BMPR1B antibody binding site. Since the number of sites is limited, as more sites are occupied by BMPR1B from the sample, fewer sites are left to bind BMPR1B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BMPR1B concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the bone morphogenetic protein (BMP) receptor family of transmembrane serine/threonine kinases. The ligands of this receptor are BMPs, which are members of the TGF-beta superfamily. BMPs are involved in endochondral bone formation and embryogenesis. These proteins transduce their signals through the formation of heteromeric complexes of 2 different types of serine (threonine) kinase receptors: type I receptors of about 50-55 kD and type II receptors of about 70-80 kD. Type II receptors bind ligands in the absence of type I receptors, but they require their respective type I receptors for signaling, whereas type I receptors require their respective type II receptors for ligand binding. Mutations in this gene have been associated with primary pulmonary hypertension. Several transcript variants encoding two different isoforms have been found for this gene.
