Intended Uses: This HIS ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey HIS. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: HIS ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-HIS antibody and an HIS-HRP conjugate. The assay sample and buffer are incubated together with HIS-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HIS concentration since HIS from samples and HIS-HRP conjugate compete for the anti-HIS antibody binding site. Since the number of sites is limited, as more sites are occupied by HIS from the sample, fewer sites are left to bind HIS-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HIS concentration in each sample is interpolated from this standard curve.

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene encodes a replication-independent member of the histone H2A family that is distinct from other members of the family. Studies in mice have shown that this particular histone is required for embryonic development and indicate that lack of functional histone H2A leads to embryonic lethality.
