Principle of the assay: MC ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MC antibody and an MC-HRP conjugate. The assay sample and buffer are incubated together with MC-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MC concentration since MC from samples and MC-HRP conjugate compete for the anti-MC antibody binding site. Since the number of sites is limited, as more sites are occupied by MC from the sample, fewer sites are left to bind MC-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MC concentration in each sample is interpolated from this standard curve.

This gene product is a chymotryptic serine proteinase that belongs to the peptidase family S1. It is expressed in mast cells and thought to function in the degradation of the extracellular matrix, the regulation of submucosal gland secretion, and the generation of vasoactive peptides. In the heart and blood vessels, this protein, rather than angiotensin converting enzyme, is largely responsible for converting angiotensin I to the vasoactive peptide angiotensin II. Angiotensin II has been implicated in blood pressure control and in the pathogenesis of hypertension, cardiac hypertrophy, and heart failure.
