This assay has high sensitivity and excellent specificity for detection of RSPO-1. No significant cross-reactivity or interference between RSPO-1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between RSPO-1 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This RSPO-1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat RSPO-1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the assay: RSPO-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-RSPO-1 antibody and an RSPO-1-HRP conjugate. The assay sample and buffer are incubated together with RSPO-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RSPO-1 concentration since RSPO-1 from samples and RSPO-1-HRP conjugate compete for the anti-RSPO-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by RSPO-1 from the sample, fewer sites are left to bind RSPO-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RSPO-1 concentration in each sample is interpolated from this standard curve.

This gene encodes a secreted activator protein with two cysteine-rich, furin-like domains and one thrombospondin type 1 domain. The encoded protein is a ligand for leucine-rich repeat-containing G-protein coupled receptors (LGR proteins) and positively regulates the Wnt signaling pathway. In mice, the protein induces the rapid onset of crypt cell proliferation and increases intestinal epithelial healing, providing a protective effect against chemotherapy-induced adverse effects. R-spondin-1: Activator of the beta-catenin signaling cascade, leading to TCF-dependent gene activation. Acts both in the canonical Wnt/beta-catenin-dependent pathway and in non-canonical Wnt signaling pathway, probably by acting as an inhibitor of ZNRF3, an important regulator of the Wnt signaling pathway. Acts as a ligand for frizzled FZD8 and LRP6. May negatively regulate the TGF-beta pathway. Has a essential roles in ovary determination. Defects in RSPO1 are the cause of palmoplantar keratoderma with squamous cell carcinoma of skin and sex reversal (PKKSCC). This recessive syndrome is characterized by XX (female to male) SRY-independent sex reversal, palmoplantar hyperkeratosis and predisposition to squamous cell carcinoma of the skin. Belongs to the R-spondin family. 3 isoforms of the human protein are produced by alternative splicing.
