Principle of the assay: ELA3B ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ELA3B antibody and an ELA3B-HRP conjugate. The assay sample and buffer are incubated together with ELA3B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ELA3B concentration since ELA3B from samples and ELA3B-HRP conjugate compete for the anti-ELA3B antibody binding site. Since the number of sites is limited, as more sites are occupied by ELA3B from the sample, fewer sites are left to bind ELA3B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ELA3B concentration in each sample is interpolated from this standard curve.

Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode the structurally similar proteins elastase 1, 2, 2A, 2B, 3A, and 3B. Unlike other elastases, elastase 3B has little elastolytic activity. Like most of the human elastases, elastase 3B is secreted from the pancreas as a zymogen and, like other serine proteases such as trypsin, chymotrypsin and kallikrein, it has a digestive function in the intestine. Elastase 3B preferentially cleaves proteins after alanine residues. Elastase 3B may also function in the intestinal transport and metabolism of cholesterol. Both elastase 3A and elastase 3B have been referred to as protease E and as elastase 1, and excretion of this protein in fecal material is frequently used as a measure of pancreatic function in clinical assays.
