specificity :
This assay has high sensitivity and excellent specificity for detection of PDL1. No significant cross-reactivity or interference between PDL1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between PDL1 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.
products description :
Intended Uses This PDL1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine PDL1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: PDL1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PDL1 antibody and an PDL1-HRP conjugate. The assay sample and buffer are incubated together with PDL1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PDL1 concentration since PDL1 from samples and PDL1-HRP conjugate compete for the anti-PDL1 antibody binding site. Since the number of sites is limited, as more sites are occupied by PDL1 from the sample, fewer sites are left to bind PDL1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PDL1 concentration in each sample is interpolated from this standard curve.

uniprot summary :
CD274: Involved in the costimulatory signal, essential for T- cell proliferation and production of IL10 and IFNG, in an IL2- dependent and a PDCD1-independent manner. Interaction with PDCD1 inhibits T-cell proliferation and cytokine production. Belongs to the immunoglobulin superfamily. BTN/MOG family. 3 isoforms of the human protein are produced by alternative splicing. Protein type: Membrane protein, integral. Chromosomal Location of Human Ortholog: 9p24. Cellular Component: integral to membrane; plasma membrane; endomembrane system; external side of plasma membrane. Molecular Function: protein binding. Biological Process: regulation of activated T cell proliferation; cell surface receptor linked signal transduction; negative regulation of interleukin-10 production; negative regulation of interferon-gamma production; negative regulation of activated T cell proliferation; T cell costimulation; positive regulation of T cell proliferation; immune response; signal transduction
