Principle of the assay: SDC-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SDC-1 antibody and an SDC-1-HRP conjugate. The assay sample and buffer are incubated together with SDC-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SDC-1 concentration since SDC-1 from samples and SDC-1-HRP conjugate compete for the anti-SDC-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by SDC-1 from the sample, fewer sites are left to bind SDC-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SDC-1 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a transmembrane (type I) heparan sulfate proteoglycan and is a member of the syndecan proteoglycan family. The syndecans mediate cell binding, cell signaling, and cytoskeletal organization and syndecan receptors are required for internalization of the HIV-1 tat protein. The syndecan-1 protein functions as an integral membrane protein and participates in cell proliferation, cell migration and cell-matrix interactions via its receptor for extracellular matrix proteins. Altered syndecan-1 expression has been detected in several different tumor types. While several transcript variants may exist for this gene, the full-length natures of only two have been described to date.
