specificity :
This assay has high sensitivity and excellent specificity for detection of IL-6. No significant cross-reactivity or interference between IL-6 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between IL-6 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This IL-6 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat IL-6. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: IL-6 ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for IL-6. Standards or samples are then added to the microtiter plate wells and IL-6 if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of IL-6 present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for IL-6 are added to each well to "sandwich" the IL-6 immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain IL-6 and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-6 concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a subunit of the interleukin 6 (IL6) receptor complex. Interleukin 6 is a potent pleiotropic cytokine that regulates cell growth and differentiation and plays an important role in the immune response. The IL6 receptor is a protein complex consisting of this protein and interleukin 6 signal transducer (IL6ST/GP130/IL6-beta), a receptor subunit also shared by many other cytokines. Dysregulated production of IL6 and this receptor are implicated in the pathogenesis of many diseases, such as multiple myeloma, autoimmune diseases and prostate cancer. Alternatively spliced transcript variants encoding distinct isoforms have been reported. A pseudogene of this gene is found on chromosome 9.[provided by RefSeq, May 2011]
uniprot summary :
IL6R: Part of the receptor for interleukin 6. Binds to IL6 with low affinity, but does not transduce a signal. Signal activation necessitate an association with IL6ST. Activation may lead to the regulation of the immune response, acute-phase reactions and hematopoiesis. Belongs to the type I cytokine receptor family. Type 3 subfamily. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Membrane protein, integral; Receptor, cytokine. Chromosomal Location of Human Ortholog: 1q21. Cellular Component: extracellular space; basolateral plasma membrane; apical plasma membrane; extracellular region; plasma membrane; interleukin-6 receptor complex. Molecular Function: protein binding; protein homodimerization activity; interleukin-6 binding; interleukin-6 receptor binding; enzyme binding; ciliary neurotrophic factor receptor activity; interleukin-6 receptor activity. Biological Process: negative regulation of collagen biosynthetic process; cytokine and chemokine mediated signaling pathway; positive regulation of smooth muscle cell proliferation; neutrophil mediated immunity; positive regulation of interleukin-6 production; positive regulation of leukocyte chemotaxis; defense response to Gram-negative bacterium; endocrine pancreas development; positive regulation of chemokine production; positive regulation of tyrosine phosphorylation of Stat3 protein; activation of NF-kappaB transcription factor; monocyte chemotaxis; positive regulation of osteoblast differentiation; defense response to Gram-positive bacterium; positive regulation of peptidyl-tyrosine phosphorylation; positive regulation of MAPKKK cascade; response to cytokine stimulus; positive regulation of cell proliferation; acute-phase response; negative regulation of interleukin-8 production; hepatic immune response.
