. Principle of the assay: MAF ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MAF antibody and an MAF-HRP conjugate. The assay sample and buffer are incubated together with MAF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MAF concentration since MAF from samples and MAF-HRP conjugate compete for the anti-MAF antibody binding site. Since the number of sites is limited, as more sites are occupied by MAF from the sample, fewer sites are left to bind MAF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MAF concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a DNA-binding, leucine zipper-containing transcription factor that acts as a homodimer or as a heterodimer. Depending on the binding site and binding partner, the encoded protein can be a transcriptional activator or repressor. This protein plays a role in the regulation of several cellular processes, including embryonic lens fiber cell development, increased T-cell susceptibility to apoptosis, and chondrocyte terminal differentiation. Defects in this gene are a cause of juvenile-onset pulverulent cataract as well as congenital cerulean cataract 4 (CCA4).
