Principle of the Assay: GAD65-Ab ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with Glutamic Acid Decarboxylase 65 antigen. Standards or samples are then added to the microtiter plate wells and GAD65-Ab if present, will bind to the antigen pre-coated wells. In order to quantitatively determine the amount of GAD65-Ab present in the sample, a standardized preparation of horseradish peroxidase (HRP) -conjugated Glutamic Acid Decarboxylase 65 antigen is added to each well to "sandwich" the GAD65-Ab immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain GAD65-Ab and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GAD65-Ab concentration in each sample is interpolated from this standard curve.

This gene encodes a cell surface receptor and transmembrane precursor protein that is cleaved by secretases to form a number of peptides. Some of these peptides are secreted and can bind to the acetyltransferase complex APBB1/TIP60 to promote transcriptional activation, while others form the protein basis of the amyloid plaques found in the brains of patients with Alzheimer disease. In addition, two of the peptides are antimicrobial peptides, having been shown to have bacteriocidal and antifungal activities. Mutations in this gene have been implicated in autosomal dominant Alzheimer disease and cerebroarterial amyloidosis (cerebral amyloid angiopathy).
