specificity :
This assay has high sensitivity and excellent specificity for detection of CTS. No significant cross-reactivity or interference between CTS and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CTS and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.
other info2 :
Intended Uses: This CTS ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey CTS. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
. Principle of the assay: CTS ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CTS antibody and an CTS-HRP conjugate. The assay sample and buffer are incubated together with CTS-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CTS concentration since CTS from samples and CTS-HRP conjugate compete for the anti-CTS antibody binding site. Since the number of sites is limited, as more sites are occupied by CTS from the sample, fewer sites are left to bind CTS-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CTS concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene, a member of the peptidase C1 family, is a lysosomal cysteine proteinase that may participate in the degradation of antigenic proteins to peptides for presentation on MHC class II molecules. The encoded protein can function as an elastase over a broad pH range in alveolar macrophages. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Dec 2010]
uniprot summary :
CTSS: Thiol protease. Key protease responsible for the removal of the invariant chain from MHC class II molecules. The bond- specificity of this proteinase is in part similar to the specificities of cathepsin L and cathepsin N. Belongs to the peptidase C1 family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Protease; EC 3.4.22.27. Chromosomal Location of Human Ortholog: 1q21. Cellular Component: extracellular space; lysosomal lumen; membrane; intracellular membrane-bound organelle; lysosome; extracellular region. Molecular Function: collagen binding; proteoglycan binding; fibronectin binding; cysteine-type endopeptidase activity; laminin binding. Biological Process: adaptive immune response; extracellular matrix organization and biogenesis; antigen processing and presentation of exogenous peptide antigen via MHC class I, TAP-independent; antigen processing and presentation of exogenous peptide antigen via MHC class II; proteolysis; antigen processing and presentation; extracellular matrix disassembly; collagen catabolic process; antigen processing and presentation of peptide antigen via MHC class I; proteolysis involved in cellular protein catabolic process; toll-like receptor signaling pathway; innate immune response; antigen processing and presentation of exogenous peptide antigen via MHC class I; immune response; positive regulation of inflammatory response
