Intended Uses: This GH ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep GH. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: GH ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-GH antibody and an GH-HRP conjugate. The assay sample and buffer are incubated together with GH-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GH concentration since GH from samples and GH-HRP conjugate compete for the anti-GH antibody binding site. Since the number of sites is limited, as more sites are occupied by GH from the sample, fewer sites are left to bind GH-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GH concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the somatotropin/prolactin family of hormones which play an important role in growth control. The gene, along with four other related genes, is located at the growth hormone locus on chromosome 17 where they are interspersed in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. The five genes share a remarkably high degree of sequence identity. Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization. This particular family member is expressed in the pituitary but not in placental tissue as is the case for the other four genes in the growth hormone locus.
