This assay has high sensitivity and excellent specificity for detection of IL-1RA. No significant cross-reactivity or interference between IL-1RA and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between IL-1RA and all the analogues, therefore, cross reaction may still exist in some cases.

Immunology

Principle of the assay: IL-1RA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-IL-1RA antibody and an IL-1RA-HRP conjugate. The assay sample and buffer are incubated together with IL-1RA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-1RA concentration since IL-1RA from samples and IL-1RA-HRP conjugate compete for the anti-IL-1RA antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-1RA from the sample, fewer sites are left to bind IL-1RA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-1RA concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the interleukin 1 cytokine family. This protein inhibits the activities of interleukin 1, alpha (IL1A) and interleukin 1, beta (IL1B), and modulates a variety of interleukin 1 related immune and inflammatory responses. This gene and five other closely related cytokine genes form a gene cluster spanning approximately 400 kb on chromosome 2. A polymorphism of this gene is reported to be associated with increased risk of osteoporotic fractures and gastric cancer. IL1RN: Inhibits the activity of interleukin-1 by binding to receptor IL1R1 and preventing its association with the coreceptor IL1RAP for signaling. Has no interleukin-1 like activity. Binds functional interleukin-1 receptor IL1R1 with greater affinity than decoy receptor IL1R2; however, the physiological relevance of the latter association is unsure. The intracellular form of IL1RN is predominantly expressed in epithelial cells. 4 isoforms of the human protein are produced by alternative splicing.
