Principle of the assay: IL-1RA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-IL-1RA antibody and an IL-1RA-HRP conjugate. The assay sample and buffer are incubated together with IL-1RA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-1RA concentration since IL-1RA from samples and IL-1RA-HRP conjugate compete for the anti-IL-1RA antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-1RA from the sample, fewer sites are left to bind IL-1RA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-1RA concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the interleukin 1 cytokine family. This protein inhibits the activities of interleukin 1, alpha (IL1A) and interleukin 1, beta (IL1B), and modulates a variety of interleukin 1 related immune and inflammatory responses. This gene and five other closely related cytokine genes form a gene cluster spanning approximately 400 kb on chromosome 2. A polymorphism of this gene is reported to be associated with increased risk of osteoporotic fractures and gastric cancer.
