This assay has high sensitivity and excellent specificity for detection of FE. No significant cross-reactivity or interference between FE and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between FE and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This FE ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey FE. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Neurobiology

Principle of the Assay: FE ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FE antibody and an FE-HRP conjugate. The assay sample and buffer are incubated together with FE-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FE concentration since FE from samples and FE-HRP conjugate compete for the anti-FE antibody binding site. Since the number of sites is limited, as more sites are occupied by FE from the sample, fewer sites are left to bind FE-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FE concentration in each sample is interpolated from this standard curve.

This gene encodes the heavy subunit of ferritin, the major intracellular iron storage protein in prokaryotes and eukaryotes. It is composed of 24 subunits of the heavy and light ferritin chains. Variation in ferritin subunit composition may affect the rates of iron uptake and release in different tissues. A major function of ferritin is the storage of iron in a soluble and nontoxic state. Defects in ferritin proteins are associated with several neurodegenerative diseases. This gene has multiple pseudogenes. Ferritin-H: Stores iron in a soluble, non-toxic, readily available form. Important for iron homeostasis. Has ferroxidase activity. Iron is taken up in the ferrous form and deposited as ferric hydroxides after oxidation. Also plays a role in delivery of iron to cells.
