Intended Uses: This TLR-2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey TLR-2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: TLR-2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TLR-2 antibody and an TLR-2-HRP conjugate. The assay sample and buffer are incubated together with TLR-2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TLR-2 concentration since TLR-2 from samples and TLR-2-HRP conjugate compete for the anti-TLR-2 antibody binding site. Since the number of sites is limited, as more sites are occupied by TLR-2 from the sample, fewer sites are left to bind TLR-2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TLR-2 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This gene is expressed most abundantly in peripheral blood leukocytes, and mediates host response to Gram-positive bacteria and yeast via stimulation of NF-kappaB. [provided by RefSeq, Jul 2008]
