specificity :
This assay has high sensitivity and excellent specificity for detection of PDGF. No significant cross-reactivity or interference between PDGF and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between PDGF and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: The sensitivity in this assay is1.0pg/mL.
other info2 :
Intended Uses: This PDGF ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep PDGF. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
products description :
Principle of the Assay: PDGF ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PDGF antibody and an PDGF-HRP conjugate. The assay sample and buffer are incubated together with PDGF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PDGF concentration since PDGF from samples and PDGF-HRP conjugate compete for the anti-PDGF antibody binding site. Since the number of sites is limited, as more sites are occupied by PDGF from the sample, fewer sites are left to bind PDGF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PDGF concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a motif of eight cysteines. This gene product can exist either as a homodimer (PDGF-BB) or as a heterodimer with the platelet-derived growth factor alpha polypeptide (PDGF-AB), where the dimers are connected by disulfide bonds. Mutations in this gene are associated with meningioma. Reciprocal translocations between chromosomes 22 and 17, at sites where this gene and that for collagen type 1, alpha 1 are located, are associated with a particular type of skin tumor called dermatofibrosarcoma protuberans resulting from unregulated expression of growth factor. Two alternatively spliced transcript variants encoding different isoforms have been identified for this gene. [provided by RefSeq, Feb 2013]
uniprot summary :
PDGFB: Growth factor that plays an essential role in the regulation of embryonic development, cell proliferation, cell migration, survival and chemotaxis. Potent mitogen for cells of mesenchymal origin. Required for normal proliferation and recruitment of pericytes and vascular smooth muscle cells in the central nervous system, skin, lung, heart and placenta. Required for normal blood vessel development, and for normal development of kidney glomeruli. Plays an important role in wound healing. Signaling is modulated by the formation of heterodimers with PDGFA. A chromosomal aberration involving PDGFB is found in dermatofibrosarcoma protuberans. Translocation t(17;22)(q22;q13) with PDGFB. Belongs to the PDGF/VEGF growth factor family. Protein type: Motility/polarity/chemotaxis; Secreted, signal peptide; Secreted; Oncoprotein. Chromosomal Location of Human Ortholog: 22q13.1.
