products description :
Principle of the Assay: YKL40 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-YKL40 antibody and an YKL40-HRP conjugate. The assay sample and buffer are incubated together with YKL40-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the YKL40 concentration since YKL40 from samples and YKL40-HRP conjugate compete for the anti-YKL40 antibody binding site. Since the number of sites is limited, as more sites are occupied by YKL40 from the sample, fewer sites are left to bind YKL40-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The YKL40 concentration in each sample is interpolated from this standard curve.

ncbi summary :
Chitinases catalyze the hydrolysis of chitin, which is an abundant glycopolymer found in insect exoskeletons and fungal cell walls. The glycoside hydrolase 18 family of chitinases includes eight human family members. This gene encodes a glycoprotein member of the glycosyl hydrolase 18 family. The protein lacks chitinase activity and is secreted by activated macrophages, chondrocytes, neutrophils and synovial cells. The protein is thought to play a role in the process of inflammation and tissue remodeling. [provided by RefSeq, Sep 2009]
uniprot summary :
CHI3L1: Carbohydrate-binding lectin with a preference for chitin. May play a role in defense against pathogens, or in tissue remodeling. May play an important role in the capacity of cells to respond to and cope with changes in their environment. A genetic variation in CHI3L1 is associated with susceptibility to asthma-related traits type 7 (ASRT7). Asthma-related traits include clinical symptoms of asthma, such as coughing, wheezing and dyspnea, bronchial hyperresponsiveness (BHR) as assessed by methacholine challenge test, serum IgE levels, atopy, and atopic dermatitis. Belongs to the glycosyl hydrolase 18 family. Protein type: Secreted; Cell adhesion; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 1q32.1. Cellular Component: proteinaceous extracellular matrix; extracellular space; endoplasmic reticulum; perinuclear region of cytoplasm; cytoplasm. Molecular Function: extracellular matrix structural constituent; chitin binding; chitinase activity; carbohydrate binding. Biological Process: positive regulation of protein kinase B signaling cascade; positive regulation of angiogenesis; apoptosis; response to mechanical stimulus; cartilage development; carbohydrate metabolic process; activation of NF-kappaB-inducing kinase; immune response; chitin catabolic process; inflammatory response; lung development. Disease: Schizophrenia; Asthma-related Traits, Susceptibility To, 7
