specificity :
This assay has high sensitivity and excellent specificity for detection of DNase B Ab. No significant cross-reactivity or interference between DNase B Ab and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between DNase B Ab and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This DNase B Ab ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human DNase B Ab. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: DNase B Ab ELISA kit applies the competitive enzyme immunoassay technique utilizing Deoxyribonuclease B antigen and an DNase B Ab-HRP conjugate. The assay sample and buffer are incubated together with DNase B Ab-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DNase B Ab concentration since DNase B Ab from samples and DNase B Ab-HRP conjugate compete for the Deoxyribonuclease B antigen binding site. Since the number of sites is limited, as more sites are occupied by DNase B Ab from the sample, fewer sites are left to bind DNase B Ab-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DNase B Ab concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene is identical to adenosine deaminase complexing protein-2, and to the T-cell activation antigen CD26. It is an intrinsic membrane glycoprotein and a serine exopeptidase that cleaves X-proline dipeptides from the N-terminus of polypeptides. [provided by RefSeq, Jul 2008]
uniprot summary :
DPP4: Cell surface glycoprotein receptor involved in the costimulatory signal essential for T-cell receptor (TCR)-mediated T-cell activation. Acts as a positive regulator of T-cell coactivation, by binding at least ADA, CAV1, IGF2R, and PTPRC. Its binding to CAV1 and CARD11 induces T-cell proliferation and NF- kappa-B activation in a T-cell receptor/CD3-dependent manner. Its interaction with ADA also regulates lymphocyte-epithelial cell adhesion. In association with FAP is involved in the pericellular proteolysis of the extracellular matrix (ECM), the migration and invasion of endothelial cells into the ECM. May be involved in the promotion of lymphatic endothelial cells adhesion, migration and tube formation. When overexpressed, enhanced cell proliferation, a process inhibited by GPC3. Acts also as a serine exopeptidase with a dipeptidyl peptidase activity that regulates various physiological processes by cleaving peptides in the circulation, including many chemokines, mitogenic growth factors, neuropeptides and peptide hormones. Removes N-terminal dipeptides sequentially from polypeptides having unsubstituted N-termini provided that the penultimate residue is proline. Belongs to the peptidase S9B family. DPPIV subfamily. Protein type: Membrane protein, integral; Protease; EC 3.4.14.5; Cell surface; Cell adhesion. Chromosomal Location of Human Ortholog: 2q24.3. Cellular Component: intercellular canaliculus; cell surface; focal adhesion; membrane; endocytic vesicle; lysosomal membrane; lamellipodium; apical plasma membrane; integral to membrane; plasma membrane; lipid raft.
