Intended Uses This HGF ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey HGF. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: HGF ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-HGF antibody and an HGF-HRP conjugate. The assay sample and buffer are incubated together with HGF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HGF concentration since HGF from samples and HGF-HRP conjugate compete for the anti-HGF antibody binding site. Since the number of sites is limited, as more sites are occupied by HGF from the sample, fewer sites are left to bind HGF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HGF concentration in each sample is interpolated from this standard curve.

Hepatocyte growth factor regulates cell growth, cell motility, and morphogenesis by activating a tyrosine kinase signaling cascade after binding to the proto-oncogenic c-Met receptor. Hepatocyte growth factor is secreted by mesenchymal cells and acts as a multi-functional cytokine on cells of mainly epithelial origin. Its ability to stimulate mitogenesis, cell motility, and matrix invasion gives it a central role in angiogenesis, tumorogenesis, and tissue regeneration. It is secreted as a single inactive polypeptide and is cleaved by serine proteases into a 69-kDa alpha-chain and 34-kDa beta-chain. A disulfide bond between the alpha and beta chains produces the active, heterodimeric molecule. The protein belongs to the plasminogen subfamily of S1 peptidases but has no detectable protease activity.
