Intended Uses: This DPP-4 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine DPP-4. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
products description :
Principle of the Assay: DPP-4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-DPP-4 antibody and an DPP-4-HRP conjugate. The assay sample and buffer are incubated together with DPP-4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DPP-4 concentration since DPP-4 from samples and DPP-4-HRP conjugate compete for the anti-DPP-4 antibody binding site. Since the number of sites is limited, as more sites are occupied by DPP-4 from the sample, fewer sites are left to bind DPP-4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DPP-4 concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene is identical to adenosine deaminase complexing protein-2, and to the T-cell activation antigen CD26. It is an intrinsic membrane glycoprotein and a serine exopeptidase that cleaves X-proline dipeptides from the N-terminus of polypeptides. [provided by RefSeq, Jul 2008]
uniprot summary :
DPP4: Cell surface glycoprotein receptor involved in the costimulatory signal essential for T-cell receptor (TCR)-mediated T-cell activation. Acts as a positive regulator of T-cell coactivation, by binding at least ADA, CAV1, IGF2R, and PTPRC. Its binding to CAV1 and CARD11 induces T-cell proliferation and NF- kappa-B activation in a T-cell receptor/CD3-dependent manner. Its interaction with ADA also regulates lymphocyte-epithelial cell adhesion. In association with FAP is involved in the pericellular proteolysis of the extracellular matrix (ECM), the migration and invasion of endothelial cells into the ECM. May be involved in the promotion of lymphatic endothelial cells adhesion, migration and tube formation. When overexpressed, enhanced cell proliferation, a process inhibited by GPC3. Acts also as a serine exopeptidase with a dipeptidyl peptidase activity that regulates various physiological processes by cleaving peptides in the circulation, including many chemokines, mitogenic growth factors, neuropeptides and peptide hormones. Removes N-terminal dipeptides sequentially from polypeptides having unsubstituted N-termini provided that the penultimate residue is proline. Belongs to the peptidase S9B family. DPPIV subfamily. Protein type: EC 3.4.14.5; Protease; Cell adhesion; Cell surface; Membrane protein, integral. Chromosomal Location of Human Ortholog: 2q24.3. Cellular Component: intercellular canaliculus; cell surface; focal adhesion; membrane; endocytic vesicle; lamellipodium; lysosomal membrane; apical plasma membrane; plasma membrane; integral to membrane; lipid raft. Molecular Function: identical protein binding; protein binding; protein homodimerization activity; protease binding; dipeptidyl-peptidase activity; serine-type peptidase activity; serine-type endopeptidase activity; receptor binding. Biological Process: regulation of cell-cell adhesion mediated by integrin; T cell activation; positive regulation of cell proliferation; T cell costimulation; response to hypoxia; endothelial cell migration; proteolysis
