Intended Uses: This FABP2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey FABP2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: FABP2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FABP2 antibody and an FABP2-HRP conjugate. The assay sample and buffer are incubated together with FABP2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FABP2 concentration since FABP2 from samples and FABP2-HRP conjugate compete for the anti-FABP2 antibody binding site. Since the number of sites is limited, as more sites are occupied by FABP2 from the sample, fewer sites are left to bind FABP2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FABP2 concentration in each sample is interpolated from this standard curve.

The intracellular fatty acid-binding proteins (FABPs) belong to a multigene family with nearly twenty identified members. FABPs are divided into at least three distinct types, namely the hepatic-, intestinal- and cardiac-type. They form 14-15 kDa proteins and are thought to participate in the uptake, intracellular metabolism and/or transport of long-chain fatty acids. They may also be responsible in the modulation of cell growth and proliferation. Intestinal fatty acid-binding protein 2 gene contains four exons and is an abundant cytosolic protein in small intestine epithelial cells. This gene has a polymorphism at codon 54 that identified an alanine-encoding allele and a threonine-encoding allele. Thr-54 protein is associated with increased fat oxidation and insulin resistance.
