specificity :
This assay has high sensitivity and excellent specificity for detection of OCLN. No significant cross-reactivity or interference between OCLN and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between OCLN and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses This OCLN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine OCLN. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: OCLN ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-OCLN antibody and an OCLN-HRP conjugate. The assay sample and buffer are incubated together with OCLN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the OCLN concentration since OCLN from samples and OCLN-HRP conjugate compete for the anti-OCLN antibody binding site. Since the number of sites is limited, as more sites are occupied by OCLN from the sample, fewer sites are left to bind OCLN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The OCLN concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes an integral membrane protein that is required for cytokine-induced regulation of the tight junction paracellular permeability barrier. Mutations in this gene are thought to be a cause of band-like calcification with simplified gyration and polymicrogyria (BLC-PMG), an autosomal recessive neurologic disorder that is also known as pseudo-TORCH syndrome. Alternative splicing results in multiple transcript variants. A related pseudogene is present 1.5 Mb downstream on the q arm of chromosome 5. [provided by RefSeq, Apr 2011]
uniprot summary :
occludin: May play a role in the formation and regulation of the tight junction (TJ) paracellular permeability barrier. It is able to induce adhesion when expressed in cells lacking tight junctions. Interacts with TJP1/ZO1 and with VAPA. Localized at tight junctions of both epithelial and endothelial cells. Highly expressed in kidney. Not detected in testis. Belongs to the ELL/occludin family. 7 isoforms of the human protein are produced by alternative splicing. Protein type: Motility/polarity/chemotaxis; Membrane protein, multi-pass; Cell adhesion; Membrane protein, integral. Chromosomal Location of Human Ortholog: 5q13.1. Cellular Component: tight junction; endocytic vesicle; apicolateral plasma membrane; apical plasma membrane; plasma membrane; integral to membrane; intercellular junction; cytoplasmic vesicle; cytosol; cell junction. Molecular Function: thiopurine S-methyltransferase activity; protein domain specific binding; protein binding; structural molecule activity. Biological Process: methylation; intercellular junction assembly and maintenance; apoptosis; S-adenosylmethionine metabolic process; S-adenosylhomocysteine metabolic process; protein complex assembly; cell structure disassembly during apoptosis. Disease: Band-like Calcification With Simplified Gyration And Polymicrogyria
