Intended Uses: This S100A7 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Goat S100A7. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: S100A7 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-S100A7 antibody and an S100A7-HRP conjugate. The assay sample and buffer are incubated together with S100A7-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the S100A7 concentration since S100A7 from samples and S100A7-HRP conjugate compete for the anti-S100A7 antibody binding site. Since the number of sites is limited, as more sites are occupied by S100A7 from the sample, fewer sites are left to bind S100A7-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The S100A7 concentration in each sample is interpolated from this standard curve.

S100A7: is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21. This protein differs from the other S100 proteins of known structure in its lack of calcium binding ability in one EF-hand at the N-terminus. This protein is markedly over-expressed in the skin lesions of psoriatic patients, but is excluded as a candidate gene for familial psoriasis susceptibility.
