products description :
Principle of the Assay: DAO ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-DAO antibody and an DAO-HRP conjugate. The assay sample and buffer are incubated together with DAO-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DAO concentration since DAO from samples and DAO-HRP conjugate compete for the anti-DAO antibody binding site. Since the number of sites is limited, as more sites are occupied by DAO from the sample, fewer sites are left to bind DAO-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DAO concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a metal-binding membrane glycoprotein that oxidatively deaminates putrescine, histamine, and related compounds. The encoded protein is inhibited by amiloride, a diuretic that acts by closing epithelial sodium ion channels. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Jan 2013]
uniprot summary :
ABP1: Catalyzes the degradation of compounds such as putrescine, histamine, spermine, and spermidine, substances involved in allergic and immune responses, cell proliferation, tissue differentiation, tumor formation, and possibly apoptosis. Placental DAO is thought to play a role in the regulation of the female reproductive function. Belongs to the copper/topaquinone oxidase family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Secreted, signal peptide; Secreted; Amino Acid Metabolism - tryptophan; EC 1.4.3.22; Oxidoreductase; Amino Acid Metabolism - histidine; Amino Acid Metabolism - arginine and proline.
