specificity :
This assay has high sensitivity and excellent specificity for detection of ECDH. No significant cross-reactivity or interference between ECDH and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ECDH and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C
other info1 :
Samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.
products description :
Intended Uses This ECDH ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken ECDH. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: ECDH ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ECDH antibody and an ECDH-HRP conjugate. The assay sample and buffer are incubated together with ECDH-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ECDH concentration since ECDH from samples and ECDH-HRP conjugate compete for the anti-ECDH antibody binding site. Since the number of sites is limited, as more sites are occupied by ECDH from the sample, fewer sites are left to bind ECDH-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ECDH concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene is a classical cadherin from the cadherin superfamily. The encoded protein is a calcium dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Mutations in this gene are correlated with gastric, breast, colorectal, thyroid and ovarian cancer. Loss of function is thought to contribute to progression in cancer by increasing proliferation, invasion, and/or metastasis. The ectodomain of this protein mediates bacterial adhesion to mammalian cells and the cytoplasmic domain is required for internalization. Identified transcript variants arise from mutation at consensus splice sites. [provided by RefSeq, Jul 2008]
uniprot summary :
CDH1: a single-pass type I membrane protein, and calcium dependent cell adhesion proteins. It is a ligand for integrin alpha-E/beta-7, and it colocalizes with DLG7 at sites of cell-cell contact in intestinal epithelial cells. Anchored to actin microfilaments through association with alpha-, beta- and gamma-catenin. Sequential proteolysis induced by apoptosis or calcium influx, results in translocation from sites of cell-cell contact to the cytoplasm. Involved in mechanisms regulating cell-cell adhesions, mobility and proliferation of epithelial cells. Defects in CDH1 are involved in dysfunction of the cell-cell adhesion system, triggering cancer invasion (gastric, breast, ovary, endometrium and thyroid) and metastasis. Has a potent invasive suppressor role. Protein type: Ubiquitin ligase; Motility/polarity/chemotaxis; Membrane protein, integral; Cell adhesion. Chromosomal Location of Human Ortholog: 16q22.1. Cellular Component: apical junction complex; internal side of plasma membrane; focal adhesion; cell surface; basolateral plasma membrane; lateral loop; integral to membrane; extracellular region; trans-Golgi network; catenin complex; actin cytoskeleton; cell-cell adherens junction; apical part of cell; perinuclear region of cytoplasm; cytoplasm; plasma membrane; nerve terminal; cell junction; lateral plasma membrane; endosome. Molecular Function: protein domain specific binding; protein binding; gamma-catenin binding; beta-catenin binding; GTPase activating protein binding; calcium ion binding; cell adhesion molecule binding; ankyrin binding; glycoprotein binding; protein phosphatase binding.
