Principle of the assay: IL-24 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-IL-24 antibody and an IL-24-HRP conjugate. The assay sample and buffer are incubated together with IL-24-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-24 concentration since IL-24 from samples and IL-24-HRP conjugate compete for the anti-IL-24 antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-24 from the sample, fewer sites are left to bind IL-24-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-24 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the IL10 family of cytokines. It was identified as a gene induced during terminal differentiation in melanoma cells. The protein encoded by this gene can induce apoptosis selectively in various cancer cells. Overexpression of this gene leads to elevated expression of several GADD family genes, which correlates with the induction of apoptosis. The phosphorylation of mitogen-activated protein kinase 14 (MAPK7/P38), and heat shock 27kDa protein 1 (HSPB2/HSP27) are found to be induced by this gene in melanoma cells, but not in normal immortal melanocytes.
