Intended Uses: This APIL ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse APIL. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: APIL ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-APIL antibody and an APIL-HRP conjugate. The assay sample and buffer are incubated together with APIL-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the APIL concentration since APIL from samples and APIL-HRP conjugate compete for the anti-APIL antibody binding site. Since the number of sites is limited, as more sites are occupied by APIL from the sample, fewer sites are left to bind APIL-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The APIL concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the tumor necrosis factor (TNF) ligand family. This protein is a ligand for TNFRSF17/BCMA, a member of the TNF receptor family. This protein and its receptor are both found to be important for B cell development. In vitro experiments suggested that this protein may be able to induce apoptosis through its interaction with other TNF receptor family proteins such as TNFRSF6/FAS and TNFRSF14/HVEM. Alternative splicing results in multiple transcript variants. Some transcripts that skip the last exon of the upstream gene (TNFSF12) and continue into the second exon of this gene have been identified; such read-through transcripts are contained in GeneID 407977, TNFSF12-TNFSF13.
