specificity :
This assay has high sensitivity and excellent specificity for detection of FE. No significant cross-reactivity or interference between FEand analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between FEand all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This FEELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine FE. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: FEELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FEantibody and an FE-HRP conjugate. The assay sample and buffer are incubated together with FE-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FEconcentration since FEfrom samples and FE-HRP conjugate compete for the anti-FEantibody binding site. Since the number of sites is limited, as more sites are occupied by FEfrom the sample, fewer sites are left to bind FE-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FEconcentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes the heavy subunit of ferritin, the major intracellular iron storage protein in prokaryotes and eukaryotes. It is composed of 24 subunits of the heavy and light ferritin chains. Variation in ferritin subunit composition may affect the rates of iron uptake and release in different tissues. A major function of ferritin is the storage of iron in a soluble and nontoxic state. Defects in ferritin proteins are associated with several neurodegenerative diseases. This gene has multiple pseudogenes. Several alternatively spliced transcript variants have been observed, but their biological validity has not been determined. [provided by RefSeq, Jul 2008]
uniprot summary :
Ferritin-H: Stores iron in a soluble, non-toxic, readily available form. Important for iron homeostasis. Has ferroxidase activity. Iron is taken up in the ferrous form and deposited as ferric hydroxides after oxidation. Also plays a role in delivery of iron to cells. Mediates iron uptake in capsule cells of the developing kidney. Oligomer of 24 subunits. There are two types of subunits: L (light) chain and H (heavy) chain. The major chain can be light or heavy, depending on the species and tissue type. The functional molecule forms a roughly spherical shell with a diameter of 12 nm and contains a central cavity into which the insoluble mineral iron core is deposited. Belongs to the ferritin family. Protein type: EC 1.16.3.1; Oxidoreductase; Cofactor and Vitamin Metabolism - porphyrin and chlorophyll. Chromosomal Location of Human Ortholog: 11q13. Cellular Component: ferritin complex; mitochondrion; cytosol; nucleus. Molecular Function: ferroxidase activity; protein binding; ferric iron binding; iron ion binding. Biological Process: receptor-mediated endocytosis; negative regulation of cell proliferation; cellular iron ion homeostasis; negative regulation of fibroblast proliferation; immune response; post-Golgi vesicle-mediated transport; transmembrane transport; iron ion transport; intracellular sequestering of iron ion. Disease: Hemochromatosis, Type 5
