Principle of the assay: ULBP2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ULBP2 antibody and an ULBP2-HRP conjugate. The assay sample and buffer are incubated together with ULBP2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ULBP2 concentration since ULBP2 from samples and ULBP2-HRP conjugate compete for the anti-ULBP2 antibody binding site. Since the number of sites is limited, as more sites are occupied by ULBP2 from the sample, fewer sites are left to bind ULBP2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ULBP2 concentration in each sample is interpolated from this standard curve.

This gene encodes a major histocompatibility complex (MHC) class I-related molecule that binds to the NKG2D receptor on natural killer (NK) cells to trigger release of multiple cytokines and chemokines that in turn contribute to the recruitment and activation of NK cells. The encoded protein undergoes further processing to generate the mature protein that is either anchored to membrane via a glycosylphosphatidylinositol moiety, or secreted. Many malignant cells secrete the encoded protein to evade immunosurveillance by NK cells. This gene is located in a cluster of multiple MHC class I-related genes on chromosome 6. Binding of ULBPs ligands to NKG2D induces calcium mobilization and activation of the JAK2, STAT5, ERK and PI3K kinase/Akt signal transduction pathway. In CMV infected cells, interacts with soluble CMV glycoprotein UL16. The interaction with UL16 blocked the interaction with the NKG2D receptor, providing a mechanism by which CMV infected cells might escape the immune system.
