specificity :
This assay has high sensitivity and excellent specificity for detection of B7-H4. No significant cross-reactivity or interference between B7-H4 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between B7-H4 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.
other info2 :
Intended Uses: This B7-H4 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse B7-H4. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
products description :
Principle of the assay: B7-H4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-B7-H4 antibody and an B7-H4-HRP conjugate. The assay sample and buffer are incubated together with B7-H4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the B7-H4 concentration since B7-H4 from samples and B7-H4-HRP conjugate compete for the anti-B7-H4 antibody binding site. Since the number of sites is limited, as more sites are occupied by B7-H4 from the sample, fewer sites are left to bind B7-H4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The B7-H4 concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a protein belonging to the B7 costimulatory protein family. Proteins in this family are present on the surface of antigen-presenting cells and interact with ligand bound to receptors on the surface of T cells. Studies have shown that high levels of the encoded protein has been correlated with tumor progression. A pseudogene of this gene is located on chromosome 20. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Dec 2011]
uniprot summary :
VTCN1: Negatively regulates T-cell-mediated immune response by inhibiting T-cell activation, proliferation, cytokine production and development of cytotoxicity. When expressed on the cell surface of tumor macrophages, plays an important role, together with regulatory T-cells (Treg), in the suppression of tumor- associated antigen-specific T-cell immunity. Involved in promoting epithelial cell transformation. Belongs to the immunoglobulin superfamily. BTN/MOG family. 3 isoforms of the human protein are produced by alternative splicing. Protein type: Membrane protein, integral. Chromosomal Location of Human Ortholog: 1p13.1. Cellular Component: integral to membrane; external side of plasma membrane. Molecular Function: receptor binding. Biological Process: response to protozoan; immune system process; positive regulation of T cell proliferation; negative regulation of T cell activation
