products description :
Intended Uses: This NTn-1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse NTn-1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: NTn-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NTn-1 antibody and an NTn-1-HRP conjugate. The assay sample and buffer are incubated together with NTn-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NTn-1 concentration since NTn-1 from samples and NTn-1-HRP conjugate compete for the anti-NTn-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by NTn-1 from the sample, fewer sites are left to bind NTn-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NTn-1 concentration in each sample is interpolated from this standard curve.

ncbi summary :
Netrin is included in a family of laminin-related secreted proteins. The function of this gene has not yet been defined; however, netrin is thought to be involved in axon guidance and cell migration during development. Mutations and loss of expression of netrin suggest that variation in netrin may be involved in cancer development. [provided by RefSeq, Jul 2008]
uniprot summary :
NTN1: Netrins control guidance of CNS commissural axons and peripheral motor axons. Its association with either DCC or some UNC5 receptors will lead to axon attraction or repulsion, respectively. It also serve as a survival factor via its association with its receptors which prevent the initiation of apoptosis. Involved in tumorigenesis by regulating apoptosis. Protein type: Secreted; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 17p13.1. Cellular Component: cytoplasm; extracellular region; basement membrane. Molecular Function: protein binding. Biological Process: axon guidance; inner ear morphogenesis; cell-cell adhesion; establishment of nucleus localization; apoptosis; positive regulation of axon extension; positive regulation of cell proliferation; neuron migration; negative regulation of axon extension; regulation of cell migration; anterior/posterior axon guidance
