Principle of the assay: DEF1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-DEF1 antibody and an DEF1-HRP conjugate. The assay sample and buffer are incubated together with DEF1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DEF1 concentration since DEF1 from samples and DEF1-HRP conjugate compete for the anti-DEF1 antibody binding site. Since the number of sites is limited, as more sites are occupied by DEF1 from the sample, fewer sites are left to bind DEF1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DEF1 concentration in each sample is interpolated from this standard curve.

Defensins are a family of microbicidal and cytotoxic peptides thought to be involved in host defense. They are abundant in the granules of neutrophils and also found in the epithelia of mucosal surfaces such as those of the intestine, respiratory tract, urinary tract, and vagina. Members of the defensin family are highly similar in protein sequence and distinguished by a conserved cysteine motif. The protein encoded by this gene, defensin, alpha 1, is found in the microbicidal granules of neutrophils and likely plays a role in phagocyte-mediated host defense. Several alpha defensin genes are clustered on chromosome 8. This gene differs from defensin, alpha 3 by only one amino acid.
