products description :
For Samples: Cell culture fluid & body fluid & tissue homogenate Serum or blood plasma. Intended Uses: This CTR ELISA kit is intended for laboratory research use only and not for use in diagnostic or therapeutic procedures. The stop solution changes the color from blue to yellow and the intensity of the color is measured at 450 nm using a spectrophotometer. In order to measure the concentration of CTR in the sample, this CTR ELISA Kit includes a set of calibration standards. The calibration standards are assayed at the same time as the samples and allow the operator to produce a standard curve of Optical Density versus CTR concentration. The concentration of in the samples is then determined by comparing the O.D. of the samples to the standard curve. Principle of the Assay: The coated well immunoenzymatic assay for the quantitative measurement of CTR utilizes a multiclonal anti-CTR antibody and an CTR-HRP conjugate. The assay sample and buffer are incubated together with CTR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CTR concentration since CTR from samples and CTR-HRP conjugate compete for the anti-CTR antibody binding site. Since the number of sites is limited, as more sites are occupied by CTR from the sample, fewer sites are left to bind CTR-HRP conjugate. Standards of known CTR concentrations are run concurrently with the samples being assayed and a standard curve is plotted relating the intensity of the color (Optical Density) to the concentration of CTR. The CTR concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a high affinity receptor for the peptide hormone calcitonin and belongs to a subfamily of seven transmembrane-spanning G protein-coupled receptors. The encoded protein is involved in maintaining calcium homeostasis and in regulating osteoclast-mediated bone resorption. Polymorphisms in this gene have been associated with variations in bone mineral density and onset of osteoporosis. Alternate splicing results in multiple transcript variants. uniprot summary :
CALCR: This is a receptor for calcitonin. The activity of this receptor is mediated by G proteins which activate adenylyl cyclase. The calcitonin receptor is thought to couple to the heterotrimeric guanosine triphosphate-binding protein that is sensitive to cholera toxin. Belongs to the G-protein coupled receptor 2 family. 3 isoforms of the human protein are produced by alternative splicing. Protein type: GPCR, family 2; Receptor, GPCR; Membrane protein, multi-pass; Membrane protein, integral. Chromosomal Location of Human Ortholog: 7q21.3. Cellular Component: integral to plasma membrane; plasma membrane. Molecular Function: protein binding; calcitonin binding; receptor activity; protein transporter activity; calcitonin receptor activity. Biological Process: G-protein coupled receptor protein signaling pathway; elevation of cytosolic calcium ion concentration; protein transport; phospholipase C activation; receptor internalization; adenylate cyclase activation; response to glucocorticoid stimulus; positive regulation of cAMP biosynthetic process; positive regulation of adenylate cyclase activity; G-protein signaling, adenylate cyclase activating pathway. Disease: Osteoporosis
