Principle of the assay: FAS/APO-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FAS/APO-1 antibody and an FAS/APO-1-HRP conjugate. The assay sample and buffer are incubated together with FAS/APO-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FAS/APO-1 concentration since FAS/APO-1 from samples and FAS/APO-1-HRP conjugate compete for the anti-FAS/APO-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by FAS/APO-1 from the sample, fewer sites are left to bind FAS/APO-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FAS/APO-1 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor contains a death domain. It has been shown to play a central role in the physiological regulation of programmed cell death, and has been implicated in the pathogenesis of various malignancies and diseases of the immune system. The interaction of this receptor with its ligand allows the formation of a death-inducing signaling complex that includes Fas-associated death domain protein (FADD), caspase 8, and caspase 10. The autoproteolytic processing of the caspases in the complex triggers a downstream caspase cascade, and leads to apoptosis. This receptor has been also shown to activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK, and is found to be involved in transducing the proliferating signals in normal diploid fibroblast and T cells. Several alternatively spliced transcript variants have been described, some of which are candidates for nonsense-mediated mRNA decay (NMD).
