Principle of the assay: GRHRAb ELISA kit applies the competitive enzyme immunoassay technique utilizing Gonadotropin Releasing Hormone Receptor antigen and an GRHRAb-HRP conjugate. The assay sample and buffer are incubated together with GRHRAb-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GRHRAb concentration since GRHRAb from samples and GRHRAb-HRP conjugate compete for the Gonadotropin Releasing Hormone Receptor antigen binding site. Since the number of sites is limited, as more sites are occupied by GRHRAb from the sample, fewer sites are left to bind GRHRAb-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GRHRAb concentration in each sample is interpolated from this standard curve.

This gene encodes the receptor for type 1 gonadotropin-releasing hormone. This receptor is a member of the seven-transmembrane, G-protein coupled receptor (GPCR) family. It is expressed on the surface of pituitary gonadotrope cells as well as lymphocytes, breast, ovary, and prostate. Following binding of gonadotropin-releasing hormone, the receptor associates with G-proteins that activate a phosphatidylinositol-calcium second messenger system. Activation of the receptor ultimately causes the release of gonadotropic luteinizing hormone (LH) and follicle stimulating hormone (FSH). Defects in this gene are a cause of hypogonadotropic hypogonadism (HH). Alternative splicing results in multiple transcript variants encoding different isoforms. Defects in GNRHR are a cause of idiopathic hypogonadotropic hypogonadism (IHH). IHH is defined as a deficiency of the pituitary secretion of follicle-stimulating hormone and luteinizing hormone, which results in the impairment of pubertal maturation and of reproductive function. Defects in GNRHR are a cause of fertile eunuch syndrome (FEUNS). Fertile eunuch syndrome is a mild phenotypic form of HH going with the presence of normal testicular size and some degree of spermatogenesis.
