specificity :
This assay has high sensitivity and excellent specificity for detection of EPO. No significant cross-reactivity or interference between EPO and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between EPO and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Assay Type: Competitive or Sandwich. Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Sensitivity: 1.0 mIU/ml.
other info2 :
Intended Uses: This EPO ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat EPO. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
products description :
Principle of the assay: EPO ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-EPO antibody and an EPO-HRP conjugate. The assay sample and buffer are incubated together with EPO-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the EPO concentration since EPO from samples and EPO-HRP conjugate compete for the anti-EPO antibody binding site. Since the number of sites is limited, as more sites are occupied by EPO from the sample, fewer sites are left to bind EPO-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The EPO concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene is a member of the EPO/TPO family and encodes a secreted, glycosylated cytokine composed of four alpha helical bundles. The protein is found in the plasma and regulates red cell production by promoting erythroid differentiation and initiating hemoglobin synthesis. This protein also has neuroprotective activity against a variety of potential brain injuries and antiapoptotic functions in several tissue types. [provided by RefSeq, Jul 2008]
uniprot summary :
EPO: Erythropoietin is the principal hormone involved in the regulation of erythrocyte differentiation and the maintenance of a physiological level of circulating erythrocyte mass. Genetic variation in EPO is associated with susceptbility to microvascular complications of diabetes type 2 (MVCD2). These are pathological conditions that develop in numerous tissues and organs as a consequence of diabetes mellitus. They include diabetic retinopathy, diabetic nephropathy leading to end-stage renal disease, and diabetic neuropathy. Diabetic retinopathy remains the major cause of new-onset blindness among diabetic adults. It is characterized by vascular permeability and increased tissue ischemia and angiogenesis. Belongs to the EPO/TPO family. Protein type: Secreted, signal peptide; Secreted. Chromosomal Location of Human Ortholog: 7q22. Cellular Component: extracellular space; cell surface; extracellular region. Molecular Function: protein binding; erythropoietin receptor binding; hormone activity; protein kinase activator activity. Biological Process: hemoglobin biosynthetic process; erythrocyte maturation; apoptosis; blood circulation; positive regulation of transcription, DNA-dependent; response to lipopolysaccharide; response to testosterone stimulus; signal transduction; response to salt stress; response to vitamin A; peptidyl-serine phosphorylation; response to hyperoxia; positive regulation of tyrosine phosphorylation of Stat5 protein; response to estrogen stimulus; positive regulation of protein kinase activity; positive regulation of cell proliferation; response to axon injury; positive regulation of Ras protein signal transduction; response to electrical stimulus; positive regulation of neuron differentiation; positive regulation of DNA replication; embryo implantation; aging.
