Principle of the assay: esRAGE ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-esRAGE antibody and an esRAGE-HRP conjugate. The assay sample and buffer are incubated together with esRAGE-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the esRAGE concentration since esRAGE from samples and esRAGE-HRP conjugate compete for the anti-esRAGE antibody binding site. Since the number of sites is limited, as more sites are occupied by esRAGE from the sample, fewer sites are left to bind esRAGE-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The esRAGE concentration in each sample is interpolated from this standard curve.

RAGE: Mediates interactions of advanced glycosylation end products (AGE). These are nonenzymatically glycosylated proteins which accumulate in vascular tissue in aging and at an accelerated rate in diabetes. Acts as a mediator of both acute and chronic vascular inflammation in conditions such as atherosclerosis and in particular as a complication of diabetes. AGE/RAGE signaling plays an important role in regulating the production/expression of TNF- alpha, oxidative stress, and endothelial dysfunction in type 2 diabetes. Interaction with S100A12 on endothelium, mononuclear phagocytes, and lymphocytes triggers cellular activation, with generation of key proinflammatory mediators. Interaction with S100B after myocardial infarction may play a role in myocyte apoptosis by activating ERK1/2 and p53/TP53 signaling. Receptor for amyloid beta peptide. Contributes to the translocation of amyloid-beta peptide (ABPP) across the cell membrane from the extracellular to the intracellular space in cortical neurons.
