specificity :
This assay has high sensitivity and excellent specificity for detection of FTH. No significant cross-reactivity or interference between FTH and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between FTH and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Assay Type: Competitive or Sandwich. Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Sensitivity: 1.0 pg/mL.
other info2 :
Intended Uses: This FTH ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human FTH. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
Priciple of the assay: FTH ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for FTH. Standards or samples are then added to the microtiter plate wells and FTH if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of FTH present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for FTH are added to each well to "sandwich" the FTH immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain FTH and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FTH concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes the heavy subunit of ferritin, the major intracellular iron storage protein in prokaryotes and eukaryotes. It is composed of 24 subunits of the heavy and light ferritin chains. Variation in ferritin subunit composition may affect the rates of iron uptake and release in different tissues. A major function of ferritin is the storage of iron in a soluble and nontoxic state. Defects in ferritin proteins are associated with several neurodegenerative diseases. This gene has multiple pseudogenes. Several alternatively spliced transcript variants have been observed, but their biological validity has not been determined. [provided by RefSeq, Jul 2008]
uniprot summary :
Ferritin-H: Stores iron in a soluble, non-toxic, readily available form. Important for iron homeostasis. Has ferroxidase activity. Iron is taken up in the ferrous form and deposited as ferric hydroxides after oxidation. Also plays a role in delivery of iron to cells. Mediates iron uptake in capsule cells of the developing kidney. Oligomer of 24 subunits. There are two types of subunits: L (light) chain and H (heavy) chain. The major chain can be light or heavy, depending on the species and tissue type. The functional molecule forms a roughly spherical shell with a diameter of 12 nm and contains a central cavity into which the insoluble mineral iron core is deposited. Belongs to the ferritin family. Protein type: Oxidoreductase; Cofactor and Vitamin Metabolism - porphyrin and chlorophyll; EC 1.16.3.1. Chromosomal Location of Human Ortholog: 11q13. Cellular Component: ferritin complex; mitochondrion; cytosol; nucleus. Molecular Function: ferroxidase activity; protein binding; ferric iron binding; iron ion binding. Biological Process: receptor-mediated endocytosis; negative regulation of cell proliferation; cellular iron ion homeostasis; negative regulation of fibroblast proliferation; immune response; post-Golgi vesicle-mediated transport; iron ion transport; transmembrane transport; intracellular sequestering of iron ion. Disease: Hemochromatosis, Type 5
