Principle of the Assay: AFP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AFP antibody and an AFP-HRP conjugate. The assay sample and buffer are incubated together with AFP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AFP concentration since AFP from samples and AFP-HRP conjugate compete for the anti-AFP antibody binding site. Since the number of sites is limited, as more sites are occupied by AFP from the sample, fewer sites are left to bind AFP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AFP concentration in each sample is interpolated from this standard curve.

This gene encodes alpha-fetoprotein, a major plasma protein produced by the yolk sac and the liver during fetal life. Alpha-fetoprotein expression in adults is often associated with hepatoma or teratoma. However, hereditary persistance of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the alpha-fetoprotein and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. Alpha-fetoprotein is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of alpha-fetoprotein in amniotic fluid is used to measure renal loss of protein to screen for spina bifida and anencephaly. [provided by RefSeq, Jul 2008]
