specificity :
This assay has high sensitivity and excellent specificity for detection of ADAM10. No significant cross-reactivity or interference between ADAM10 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ADAM10 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This ADAM10 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse ADAM10. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: ADAM10 ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for ADAM10. Standards or samples are then added to the microtiter plate wells and ADAM10 if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of ADAM10 present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for ADAM10 are added to each well to "sandwich" the ADAM10 immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain ADAM10 and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADAM10 concentration in each sample is interpolated from this standard curve.

ncbi summary :
Members of the ADAM family are cell surface proteins with a unique structure possessing both potential adhesion and protease domains. This gene encodes and ADAM family member that cleaves many proteins including TNF-alpha and E-cadherin. [provided by RefSeq, Jul 2008]
uniprot summary :
ADAM10: Cleaves the membrane-bound precursor of TNF-alpha at 76-Ala- -Val-77 to its mature soluble form. Responsible for the proteolytical release of soluble JAM3 from endothelial cells surface. Responsible for the proteolytic release of several other cell-surface proteins, including heparin-binding epidermal growth- like factor, ephrin-A2 and for constitutive and regulated alpha- secretase cleavage of amyloid precursor protein (APP). Contributes to the normal cleavage of the cellular prion protein. Involved in the cleavage of the adhesion molecule L1 at the cell surface and in released membrane vesicles, suggesting a vesicle-based protease activity. Controls also the proteolytic processing of Notch and mediates lateral inhibition during neurogenesis. Responsible for the FasL ectodomain shedding and for the generation of the remnant ADAM10-processed FasL (FasL APL) transmembrane form. Also cleaves the ectodomain of the integral membrane proteins CORIN and ITM2B. May regulate the EFNA5-EPHA3 signaling. Protein type: Cell surface; Membrane protein, integral; EC 3.4.24.81; Cell adhesion; Vesicle; Protease; Motility/polarity/chemotaxis. Chromosomal Location of Human Ortholog: 15q22.
