specificity :
This assay has high sensitivity and excellent specificity for detection of Ang2. No significant cross-reactivity or interference between Ang2 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between Ang2 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 1.0 pg/mL.
other info2 :
Intended Uses: This Ang2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse Ang2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
Principle of the assay: Ang2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Ang2 antibody and an Ang2-HRP conjugate. The assay sample and buffer are incubated together with Ang2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Ang2 concentration since Ang2 from samples and Ang2-HRP conjugate compete for the anti-Ang2 antibody binding site. Since the number of sites is limited, as more sites are occupied by Ang2 from the sample, fewer sites are left to bind Ang2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Ang2 concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene is an antagonist of angiopoietin 1 (ANGPT1) and endothelial TEK tyrosine kinase (TIE-2, TEK). The encoded protein disrupts the vascular remodeling ability of ANGPT1 and may induce endothelial cell apoptosis. Three transcript variants encoding three different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
uniprot summary :
ANGPT2: Binds to TEK/TIE2, competing for the ANGPT1 binding site, and modulating ANGPT1 signaling. Can induce tyrosine phosphorylation of TEK/TIE2 in the absence of ANGPT1. In the absence of angiogenic inducers, such as VEGF, ANGPT2-mediated loosening of cell-matrix contacts may induce endothelial cell apoptosis with consequent vascular regression. In concert with VEGF, it may facilitate endothelial cell migration and proliferation, thus serving as a permissive angiogenic signal. Interacts with TEK/TIE2, competing for the same binding site as ANGPT1. 3 isoforms of the human protein are produced by alternative splicing. Protein type: Secreted; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 8p23.1. Cellular Component: extracellular space; cell projection; extracellular region; plasma membrane; nucleus.
