Principle of the assay: CRY-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CRY-1 antibody and an CRY-1-HRP conjugate. The assay sample and buffer are incubated together with CRY-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CRY-1 concentration since CRY-1 from samples and CRY-1-HRP conjugate compete for the anti-CRY-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by CRY-1 from the sample, fewer sites are left to bind CRY-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CRY-1 concentration in each sample is interpolated from this standard curve.

The photoreceptor may be involved in electron transport. Mutant phenotype displays a blue light-dependent inhibition of hypocotyl elongation. Photoreceptor activity requires light-induced homodimerisation of the N-terminal CNT1 domains of CRY1. Involved in blue-light induced stomatal opening. The C-terminal domain of the protein undergoes a light dependent conformational change. Also involved in response to circadian rhythm. Mutants exhibit long hypocotyl under blue light and are out of phase in their response to circadian rhythm. CRY1 is present in the nucleus and cytoplasm. Different subcellular pools of CRY1 have different functions during photomorphogenesis of Arabidopsis seedlings.
